🔬 Cryoprotectant exposure
Cryoprotectants (CPAs) such as ethylene glycol and DMSO help reduce intracellular ice formation.
Insufficient CPA penetration may increase the risk of ice formation. Excessive exposure can cause chemical toxicity and osmotic stress.

💧 Osmotic changes
When an embryo enters a solution containing a non-permeating solute, water moves out of its cells by osmosis, causing shrinkage.
Permeating CPAs enter the cells, changing intracellular and extracellular osmotic balance.
During subsequent steps, water re-enters the cells.
Excessive shrinkage or dwelling can damage cellular membranes and organelles.

❄️ Ice crystal formation
Vitrification aims to convert the solution into a glass-like, non-crystalline state.
Ice crystals can damage cell membranes, organelles and intracellular structures.
High cooling rates, adequate CPA concentrations and minimal solution volume help reduce ice formation.

🌡️ Cooling rate and warming rate
Rapid cooling reduces the opportunity for ice crystals to nucleate and grow.
Warming rate is equally critical. Slow warming can allow ice crystals to form or grow through devitrification and recrystallization.

🧬 Embryo stage and cellular structure
Blastocysts: The blastocoel contains fluid, and its volume can influence vitrification outcomes.
Cleavage-stage embryos: Blastomere membranes and intracellular structures must be protected from osmotic and cryogenic injury.
Embryos with different stages or characteristics may respond differently to cryopreservation.

🧪 Residual solution volume
A minimal, appropriate volume around the embryo helps achieve rapid cooling.
Excess solution can reduce the effective cooling rate and increase the amount of solution that must vitrify.
Avoid compromising embryo retention or allowing the embryo to dry.

✨Devitrification and recrystallization
Devitrification: A glass-like vitrified state begins forming crystals during warming.
Recrystallization: Small ice crystals grow or reorganize into larger crystals.
Both can injure the embryo, which is why the prescribed warming procedure is crucial.

The ICSI dish is designed to create a controlled, organized workspace for micromanipulation. It provides separate drops for oocyte handling and sperm.

In an ICSI dish, PVP and HEPES-buffered handling medium have different jobs.

✨PVP (polyvinylpyrrolidone) is mainly used for sperm handling and immobilization

✨It increases the viscosity around the sperm, making individual sperm easier to pick up with the injection pipette.

✨It helps slow sperm movement, allowing selection of a suitable sperm.

✨It helps the sperm remain relatively stable while loading it into the injection pipette.

✨It reduces the tendency of sperm to move away from the pipette during manipulation.

✨PVP is not injected intentionally as a medium into the oocyte. Only a minimal amount should accompany the sperm

🌸HEPES-buffered medium is generally used for oocyte handling outside the CO₂ incubator.

🌸HEPES acts as a pH buffer, helping maintain a suitable pH while the dish is exposed to atmospheric air rather than the controlled CO₂ environment of the incubator.

❄️Carefully overlay the drops with pre-equilibrated sterile mineral oil.

❄️Oil prevents evaporation, helps maintain osmolality and pH, and provides a stable environment during manipulation

📍Holding pipette → oocyte stabilization.
📍Injection pipette → sperm immobilization and injection.

In some way, we are part of deciding which oocyte 🥚 and sperm will come together. From a microscopic world that no one can see with the naked eyes 👁️, we help create the beginning of a new life..🐣💞

A new life, a new story, a new family...all starting from a few microscopic cells and the careful work of our hands🙌🌼....Every step requires patience, care, and responsibility. We watch, choose, handle, and protect these tiny beginnings, knowing that someday they may become someone’s whole world. 🥹✨

So Maybe we can call as our magical hands 🧚‍♀️🪄🔬

Wishing every embryologist’s hands to always be gentle, skilled, and magical enough to give these tiny beginnings their best chance to become human beings, experience life, and see the beautiful world beyond the microscope. 💞🧫✨

👀 You’re looking at a cell… but do you know how special it is? 🥚🔬

📍The largest human cell! 👑
A mature oocyte is about 100–120 µm in diameter—huge compared with most human cells.

📍Born before you are born! 👶🏻
Most oocytes are formed during fetal development, before birth.

📍It has its own protective layers 🛡️
The oocyte is surrounded by the zona pellucida and corona radiata, which play important roles in fertilization and protection.

📍The oocyte contributes more than DNA 🧬
At fertilization, the oocyte provides not only its nuclear genome but also cytoplasm, organelles, maternal RNAs, proteins, and mitochondria.

📍The first polar body isn't “waste” 👀
It is a by-product of unequal meiotic division and can be an important marker of oocyte maturity in the IVF lab.

📍⏳ Age affects oocyte quality, not simply the number of oocytes. Chromosomal errors become more common with increasing maternal age

📍It can wait for decades! ⏳
Primary oocytes can remain arrested in prophase I for many years until they resume maturation.

📍It changes dramatically after fertilization! 🔬
Following sperm entry, the oocyte activates, completes meiosis II, and forms the female pronucleus.

📍The oocyte contributes more than DNA 🧬
At fertilization, the oocyte provides not only its nuclear genome but also cytoplasm, organelles, maternal RNAs, proteins, and mitochondria.

📍In IVF: Embryologists assess maturity, morphology, cytoplasmic appearance, and integrity before procedures such as ICSI.

📍Maturity stages: Commonly classified as GV (germinal vesicle), MI (metaphase I), and MII (metaphase II).

🔬 Important points to remember

Check media: expiry date, lot number, storage condition and appearance before use.

Equilibration: bicarbonate-buffered media should be properly equilibrated in the incubator.

Temperature: avoid exposing oocytes to temperature fluctuations; use a validated heated surface where required.

pH/osmolality: don't leave bicarbonate-buffered media outside controlled conditions longer than necessary.

Dish labelling: clearly identify patient/case ID, date and relevant dish details according to your SOP.

Avoid evaporation: prepare appropriate drops/volumes and maintain adequate oil coverage.

Aseptic technique: use sterile consumables and avoid touching the inside of wells/drops with pipette tips.

Documentation: record media lot numbers and other required details.

🥚 Denudation is not just “removing cells.” It allows accurate assessment of the oocyte’s maturity before ICSI.

🧪 Hyaluronidase does not remove all cumulus cells by itself. Mechanical pipetting is essential for complete denudation.

⏱️ More enzyme exposure is not better. The aim is to use the minimum exposure required according to the validated SOP.

🔬 Pipette diameter matters. A larger pipette can remove bulk cumulus cells, while a smaller one helps achieve finer denudation.

⚠️ Overly forceful pipetting can damage the oocyte, even when the oocyte looks normal externally.

👀 Denudation reveals the first polar body, which is an important indicator for identifying an MII oocyte.

🧬 Not every retrieved oocyte is mature. Denudation helps distinguish GV, MI and MII stages.

🥚 The corona radiata is closely associated with the oocyte, so complete removal requires careful mechanical handling.

🌡️ Temperature and pH matter during denudation. Oocytes should spend as little unnecessary time outside controlled culture conditions as possible.

🧼 Washing after denudation is important to remove residual enzyme and cellular debris.

🔍 Denudation also makes morphological assessment easier, including evaluation of the ooplasm and perivitelline space.

🎯 Good denudation = clean enough for assessment and ICSI, but gentle enough to preserve oocyte integrity. by @_embryo_scope_
