Could Thymosin-α1 have future research potential in fibromyalgia? 🧠🧬

Preclinical studies suggest Tα1 may influence pain signalling, microglial activation and inflammatory pathways that overlap with mechanisms being investigated in fibromyalgia.

However, there are currently no published human trials of Tα1 specifically for fibromyalgia, so this remains an experimental research hypothesis rather than an established treatment.

🔬 Interesting biology. Promising questions. More research needed.

5-Amino-1MQ is a small-molecule NNMT inhibitor studied for its potential effects on cellular metabolism and adipose tissue.

In the preclinical study by Neelakantan et al., published in Biochemical Pharmacology (2018), 5-Amino-1MQ reduced intracellular 1-MNA, increased NAD⁺ and SAM in cultured adipocytes, suppressed lipogenesis, and was associated with lower body weight, adipose mass and total cholesterol in diet-induced obese mice.

Every heartbeat needs energy🫀

Inside heart muscle cells, mitochondria help produce the energy that keeps the heart pumping. SS-31 is a small peptide studied for its effects on these cellular energy producers.

In mouse studies published in 2017 and 2022, researchers made the heart pump against greater forced resistance. Compared with untreated mice, those given SS-31 showed:

• Less abnormal heart enlargement
• Less scarring (fibrosis)
• Better pumping function

Together, these findings suggest that protecting mitochondria may help limit heart damage from pressure overload. These animal and laboratory findings do not establish benefits or safety for people with heart disease.

Swipe through to explore the research. The final slide features our Janoshik COA, reporting 99.689% purity and 50.96 mg SS-31 in the tested sample.

Tesamorelin works upstream in the GH/IGF-1 axis.

Rather than supplying growth hormone directly, tesamorelin is a GHRH analogue studied for its interaction with GHRH receptors at the pituitary promoting endogenous GH signaling and, downstream, IGF-1 production.

A simple way to think about it:

Tesamorelin → pituitary signaling → endogenous GH → liver → IGF-1

Same pathway, different entry point.

Understanding where a compound acts within a signaling pathway is essential when interpreting research data.

‘MOTS-c reduces myostatin and muscle atrophy signaling’ 🧬📑

A 2021 study explored how MOTS-c, a mitochondrial-derived peptide, may influence myostatin and muscle atrophy signaling.

Researchers reported that:

• Higher circulating MOTS-c was associated with lower myostatin levels in humans
• In muscle cells, MOTS-c helped protect against palmitic-acid-induced atrophy-like changes
• In high-fat-diet mice, MOTS-c reduced circulating myostatin by around 40%
• Skeletal-muscle myostatin expression also decreased
• The proposed mechanism involved the CK2 → PTEN → mTORC2 → AKT → FOXO1 pathway
• In a short-term mouse model, MOTS-c helped preserve relative muscle mass under high-fat-diet conditions

The interesting part? Myostatin is one of the body’s key regulators of muscle growth and muscle-wasting signaling, making this pathway particularly relevant to research into sarcopenia and metabolic muscle dysfunction.

Important: this was not a human clinical trial testing MOTS-c as a treatment for muscle loss. The evidence came from human correlation data, cell experiments and animal models.

AOD-9604 vs hGH-Frag 176-191 vs full hGH 🧬

They’re related - but they’re definitely not the same molecule.

🔹 AOD-9604 is a modified peptide derived from the C-terminal lipolytic region of hGH, designed to investigate metabolic/fat-related effects without reproducing the full hormonal activity of growth hormone.

🔹 hGH-Frag 176-191 is the native C-terminal fragment of human growth hormone and has also been investigated mainly for its potential lipolytic activity.

🔹 Full hGH (1-191) is the complete 191-amino-acid hormone, with much broader endocrine effects- including GH receptor activation, increased IGF-1, anabolic signaling, effects on glucose metabolism, and feedback suppression of endogenous GH release.

One important molecular detail: AOD-9604 and hGH-Frag 176-191 come from essentially the same C-terminal region, but their sequences are not identical. AOD-9604 uses an N-terminal Tyrosine (Y) modification, while the native fragment begins with Phenylalanine (F) keeping the native hGH portion associated with lipolysis.

Same family. Very different biological profiles. 🔬

What if a mitochondrial peptide could help protect the brain from sleep deprivation? 🧠⚡️

SS-31 (also known as elamipretide) has been investigated in preclinical studies for its potential to reduce some of the cognitive and mitochondrial effects linked to sleep loss.

Research in mice has associated SS-31 with:
• better learning and memory performance
• reduced oxidative stress
• lower inflammatory markers
• improved mitochondrial function

That said, the evidence is still preclinical, and more human research is needed.

What if protecting mitochondria could also help protect vision? 👁️⚡

A recent in-vivo study in an experimental glaucoma model explored whether SS-31 (Elamipretide) could help protect retinal ganglion cells - the cells responsible for transmitting visual information from the eye to the brain.

Researchers observed that SS-31 was associated with:

👁️ Greater retinal ganglion cell survival
🧬 Better preservation of retinal structure
⚡ Improved visual responses
🔋 Potential protection against mitochondrial dysfunction and oxidative stress

Because retinal ganglion cells are highly dependent on healthy mitochondrial function, these findings raise an interesting question: could mitochondria-targeted peptides like SS-31 eventually contribute to new strategies for retinal diseases?

Important: this research was conducted in an animal model, not in humans. It does not establish SS-31 as a treatment for retinal disease and/or glaucoma.

What if MOTS-c could be a potential treatment/adjuvant for osteoporosis? 🤔

A 2019 in-vitro study using human osteoblast cells explored an interesting mechanism linking MOTS-c to bone biology.

Researchers observed that MOTS-c:

🦴 Increased osteoblast viability
🧬 Increased expression of COL1A1 and COL1A2, genes involved in Type I collagen production
🔬 Increased corresponding collagen-related protein expression
⚙️ Appeared to act, at least partly, through the TGF-β/SMAD signaling pathway

Type I collagen represents around 80–90% of bone’s organic matrix, making its synthesis particularly relevant to bone structure and osteoporosis research.

The findings raise an interesting research question: could MOTS-c eventually contribute to new approaches for supporting bone formation in osteoporosis?

Important: this was a cell-based study, not a human clinical trial. It does not demonstrate that MOTS-c treats osteoporosis, establish human dosing, or confirm clinical safety or efficacy.

The Origins of Semax 🧠

Semax traces its origins back to Soviet peptide research in the late 1970s and 1980s, when scientists were investigating short fragments of ACTH (adrenocorticotropic hormone) and their effects on the nervous system.

Researchers at the Institute of Molecular Genetics and Moscow State University focused on the ACTH(4–7) fragment and added a Pro-Gly-Pro (PGP) sequence to improve its stability. The result was the seven-amino-acid peptide now known as Semax: Met-Glu-His-Phe-Pro-Gly-Pro (MEHFPGP). (Sparta Labs)

What began as a Soviet-era neuropeptide research programme would later develop into decades of research exploring Semax in areas such as neuroprotection, cognition and nervous-system function.

A small peptide with a surprisingly long research history.
