How has Down syndrome shaped Alzheimer's research?
Nearly 40 years ago, people with Down syndrome helped researchers discover the gene behind Alzheimer's abnormal protein buildup. Now ATRI is bringing Alzheimer's research back to the community that helped found it. The USC Epstein Family ATRI has built a large consortium of sites for Down syndrome research and plans on launching two Down syndrome-related-Alzheimer's disease studies this upcoming year.

It's easy to assume that clinical trials only come into play after receiving an Alzheimer's Disease diagnosis. But what many people don't realize is that a growing number of trials are focused on something different: preventing and delaying the onset of Alzheimer's Disease altogether. These prevention-focused studies represent a crucial shift in how we approach the disease: Rather than waiting to treat symptoms, they aim to intervene earlier, before cognitive decline begins.
Interested in preventative Alzheimer's Disease trials? Check out the Alzheimer's Prevention Trials (APT) Webstudy - A fully virtual study that lets participants monitor their own cognitive health over time, while giving eligible individuals the opportunity to fast-track into other clinical trials. USC hosted a special celebration for the historic renaming of the Epstein Family Alzheimer's Therapeutic Research Institute.

The Epstein family's generosity and focus are accelerating the path to Alzheimer’s prevention. They are solidifying USC's role as the leader in Alzheimer's and related dementias research.

Could taking care of your overall health also help protect your brain?

USC researchers are exploring how factors such as physical activity, cardiovascular health and the environment may influence the risk of developing Alzheimer’s disease.

In the third installment of USC United Against Alzheimer’s: A Collaborative Approach to Solving Alzheimer’s, learn how researchers are looking beyond the brain to better understand Alzheimer’s risk and the role overall health may play. Read here: https://uscne.ws/aoh

New Waves of Research from ATRI connect the link between Down syndrome and Alzheimer’s disease
The Union Tribune published an article highlighting the work of USC Epstein Family ATRI and Dr. Michael Rafii in the field of Down syndrome research. For the past 15 year, Dr. Rafii and his team have studied the relationship between Alzheimer’s disease and Down syndrome. Additionally, how to include people with Down syndrome in Alzheimer’s research in a supportive and inclusive environment, such as projects like Noah Homes or ATRI’s Down syndrome ambassadors at research sites.
“Our goal is not simply to understand why Alzheimer’s disease occurs in people with Down syndrome, but to develop treatments that can delay, prevent, or effectively treat it and ultimately give people with Down syndrome the opportunity to live longer, healthier lives without dementia” – Dr. Michael Rafii

Progress should not stay in one lab or institution.
The new FDA approved Roche and Eli Lilly's pTau217 blood test for Alzheimer's disease, the first single-biomarker immunoassay, will allow labs without mass spectrometers to run Alzheimer’s Disease blood tests.

Unlike other tests, the pTau217 immunoassay looks solely at pTau217 levels, rather than ratios with other proteins. The immunoassay can run on standard instruments already in commercial labs nationwide in about 18 minutes, while mass spectrometry tests require specialized lab equipment. This means faster, more accessible answers for patients and providers everywhere, not just those near specialized testing centers. Both LapCorp and Quest Diagnostics are currently planning on offering the pTau217 blood test.

At USC's Epstein Family ATRI, we're proud to play a role in this progress. Our team is continually accelerating what we know about Alzheimer’s and related dementias. In our labs, we have the mass spectrometry equipment and immunoassay expertise to develop and verify new biomarker tests and the largest dementia plasma biorepository in the U.S. We share that data with scientists everywhere, because progress on Alzheimer's shouldn't stay in one lab. Progress in Alzheimer’s Disease and Related Dementia research requires exchanging ideas and information, and we believe that collaboration is paramount to move the field forward. by @atriusc

Congratulations to our very own Dr. Charisse Winston on receiving an Alzheimer's Association Research Grant (AARG) for her project: "Saliva Extracellular Vesicles as Alzheimer's Disease Diagnostic Biomarkers".
Dr. Winston's project ACCELERATES the field of biofluid-based biomarkers by establishing salivary extracellular vesicles as a groundbreaking, noninvasive diagnostic platform for Alzheimer's Disease.
"By translating complex molecular profiling to an easily accessible biofluid, this work breaks through traditional logistical and infrastructure barriers, laying the foundation for scalable, equitable, and cost-effective screening tools that enable early, timely diagnosis across diverse and historically underserved populations." - Dr. Charisse Winston by @atriusc

By the Numbers:
Clinical trials are an essential piece in the roadmap to prevention. Yet, 37% of clinical trials do not have enough participants and 11% of clinical trials are unable to enroll any participants.

Libby Sofar was dementia patient in hospice when, like many dementia patients, she experienced agitation and confusion. She received standard hospice end-of-life therapies such as Haloperidol, Valium, and Morphine. This combination only increased her agitation, confusion, and despair.
The USC Epstein Family ATRI team, along with the Alzheimer's Clinical Trials Consortium (ACTC), developed LiBBY to find a better end-of-life treatment for dementia patients by using THC and CBD. Study results show that THC and CBD had a statistically significant positive effect on behavior and agitation.
